چکیده

Twenty-five strains of Fusarium sambucinum grown on wheat kernels were examined for trichothecene production and the synthesis of volatile sesquiterpenes. The volatiles were purged with air and collected on Tenax traps. Adsorbed compounds were eluted from the traps and injected into a gas chromatograph coupled with a mass spectrometer. Ten strains isolated from potato tubers produced high amounts of diacetoxyscirpenol and its derivatives. These strains were characterized by the production of high amounts of diverse sesquiterpenes. In 10 cultures, 19 compounds were detected, of which 6 were predominant and composed as much as 82% of the volatile sesquiterpene fraction (e.g., beta-farnesene, beta-chamigrene, beta-bisabolene, alpha-farnesene, trichodiene, and an unidentified compound). Fifteen strains isolated from various sources that did not produce trichothecenes produced much less volatile sesquiterpenes, with less chemical diversity. No more than six compounds were present in cultures. Two of these compounds were present in the toxigenic strains isolated from potatoes (beta-farnesene and acoradiene), but four were unique to the strains not producing trichothecenes (longifolene, isocaryophyllene, delta-elemene, and an unidentified one). The pattern of volatile sesquiterpenes was characteristic and distinctive for both toxic and nontoxic strains.

A Fungal Symbiont of Plant-Roots Modulates Mycotoxin Gene Expression in the Pathogen Fusarium sambucinum

Fusarium trichothecenes are fungal toxins that cause disease on infected plants and, more importantly, health problems for humans and animals that consume infected fruits or vegetables. Unfortunately, there are few methods for controlling mycotoxin production by fungal pathogens. In this study, we isolated and characterized sixteen Fusarium strains from naturally infected potato plants in the f...

The arbuscular mycorrhizal fungus, Glomus irregulare, controls the mycotoxin production of Fusarium sambucinum in the pathogenesis of potato.

Trichothecenes are an important family of mycotoxins produced by several species of the genus Fusarium. These fungi cause serious disease on infected plants and postharvest storage of crops, and the toxins can cause health problems for humans and animals. Unfortunately, there are few methods for controlling mycotoxin production by fungal pathogens, and most rely on chemicals, creating therefore...

Clustering of trichothecene-producing Fusarium strains determined from 28S ribosomal DNA sequences.

The genus Fusarium includes several species that produce trichothecenes. We analyzed DNA sequences from a variable region at the 5' end of the large nuclear ribosomal DNA (rDNA) (28S) to determine the genetic relatedness of trichothecene-producing Fusarium species. All trichothecene-producing strains clustered together, and two monophyletic groups were resolved. The first clade included strains...

Myrothecium roridum Tri4 encodes a multifunctional oxygenase required for three oxygenation steps.

The biosyntheses of both macrocyclic trichothecenes in Myrothecium roridum and simple trichothecenes in Fusarium species begin with the cyclization of farnesyl pyrophosphate to form the sesquiterpene hydrocarbon trichodiene. A previous study showed that Myrothecium has a cluster of 3 genes that are homologous with Fusarium trichothecene genes: Tri4, a P450 oxygenase; Tri5, the sesquiterpene cyc...

New modified trichothecenes accumulated in solid culture by mutant strains of Fusarium sporotrichioides.

Mutant strains of Fusarium sporotrichioides NRRL 3299 deficient in the ability to synthesize T-2 toxin were examined on solid rice medium. Five novel alicyclic trichothecenes were isolated: 11 alpha-hydroxytrichodiene; tricho-9-ene-2 alpha,3 alpha,11 alpha-triol; tricho-9-ene-2 alpha,3 alpha,8 alpha,11 alpha-tetraol; tricho-9-ene-2 alpha,3 alpha,8 beta,11 alpha-tetraol; and tricho-9-ene-2 alpha...
